چکیده

Abstract Study question Which sperm preparation procedure is able to select the best population of sperm? Summary answer Microfluidic sorting using Fertile Plus device isolates with highest motility and lowest levels DNA fragmentation. What known already The selection in Medically Assisted Reproduction should lead fully functional spermatozoa reach fertilization site allow optimal embryo development. These abilities are dependent on specific parameters such as, but not limited to, motility, morphology, Acrosome Index (AI), fragmentation (DFI). At present, routine suboptimal, does replicates physiological processes observed nature consequently may induce damage. new Sperm Sorting technology allows a sperm. However, no comparison other procedures regarding all above-mentioned was reported. design, size, duration Prospective-observational study performed at an University Hospital-IVF Centre between April-November 2022 comparing effect four different various semen parameters. Fifty-two samples minimum 20x106 sperm/ml progressive ≥30% were included. Descriptive statistics reported mean±SD [minimum-maximum] values for each variable. Interclass correlation coefficient (ICC) calculated absolute agreement procedures, where DGC considered standard-of-care. An ICC-value <0.4 indicates low agreement. Participants/materials, setting, methods Unused raw after diagnostic analysis individually split 4 fractions processed immediately by methods: 1) wash (SW), 2) Density Gradient Centrifugation (DGC), 3) Magnetic Activated Cell (MACS, (Miltenyi Biotec)), 4) (MSS) Device (Koek Biotechnology). Each fraction analyzed (AI) (DFI; TUNEL assay protocol). Main results role chance mean age patients 35.9 years (SD ± 6.8). value days abstinence 4.1 Mean rates were: 54.3±10.6% [23-86%], 74.3±11.8% [38-90%], 77.2±12.5% [37-92%] 88.6±4.2% [73-96%] SW, DGC, MACS, MSS, respectively. percentages normal morphology 3.3±2.9% [0-13%], 4.1±3.1% 4.2±3.7% [0-18%], 5.1±3.9% [0-16%] AI 8.5±4.9% [1-20%], 9.7±6.0% [1-30%], 8.7±4.9% [0-19%], 10.8±6.8% [1-30%] DFI 6.2±4.6% [0.8-26.1%], 2.7±3.2% [ 0.2-14%], 2.1±4.3% [0.9-20.8%], 0.2±0.4% [0.0-2.3%] Values referring ICC indicate MSS as only strong disagreement [0.29, 95%CI (-0.2-0.58)] [0.17; (-0.19-0.45)]. No techniques. A further paired confirmed MACS [0.2; (-0.17-0.49) 0.21; (-0.25-0.52)] SW [0.06; (-0.04-0.25) 0.06; (-0.15-0.28)] DFI, Limitations, reasons caution Differences DNA-damage have been smaller techniques if period shorter. Due design experiment, data clinical outcomes available. Wider implications findings highly motile undamaged from unprocessed ideally Plus. Question remains how these compare shorter whether this improves embryological IVF laboratory. Trial registration number NCT05383599

P-240: Effect of Oxidative Stress on DNA Fragmentation Index in Sperm of Mature Mice

Background: One of the major factors affecting on male infertility is oxidative stress (OS). Two main sources of ROS production are leukocytes and spermatozoa. The ROS such as tertiary-butyl hydroperoxide (TBHP) may damage and fragmentate to sperm DNA, which can lead to complications in the offspring. In this study, we will investigate the effect of od on DNA fragmentation index (DFI) in sperm ...

Sperm aneuploidies and low progressive motility.

BACKGROUND Patients with poor semen quality show increased sperm disomy and diploidy rates. Oligozoospermia and teratozoospermia are known to influence sperm aneuploidy, but there is still a debate about whether aneuploidies are associated with reduced motility. METHODS Ejaculates from a large group of patients were examined by light microscopy to evaluate sperm concentration, motility and mo...

P-13: Comparison of Sperm Quality, Oxidative Stress, DNA Fragmentation, Protamine Deficiency, and DNA Methylation in Varicocele and Fertile Individuals

s:1993:"Background: There are many approaches that gene expression is controlled in eukaryotes. DNA methylation is one of several epigenetic mechanisms that cells use to control gene expression and lock genes in the "off" position. In addition, sperm DNA damage can correlate with DNA methylation defect. There is evidence that sperm of infertile men contain more DNA damage than fertile men and t...

P-23: Evaluation of Relation between Sperm DNA Fragmentation with IUI Outcomes

Background: Sperm chromatin integrity assessment has been suggested as fertility predictor. The aim of this study was to examine the relationship between the results of sperm chromatin structure assay (SCSA) and the outcome of intrauterine insemination IUI. Materials and Methods: A total of 576 consecutive couples undergoing IUI were included and IUI was performed. SCSA results were expressed a...

Is sperm DNA fragmentation a good

This paper aimed at investigating the potential use of sperm DNA fragmentation (SDF) to improve the routine screening of infertility of Holstein bulls. Cryopreserved sperm samples from 201 Holstein bulls provided by an AI center were used in the analyses of SDF at 0 (SDF_0) and 6 (SDF_6) h of incubation at 37oC. A refi nement of the sperm chromatin dispersion test implemented in the Sperm-Halom...
